Production of Recombinant Streptokinase
Introduction
Streptokinase, a thrombolytic drug,
of great therapeutic value is produced commercially
by E.coli. The method used by us depends on Bacillus
Subtilis, which is a GRAS (Generally Regarded as Safe)
bacteria. The drug will be produced by inserting gene
for streptokinase in a high copy no. plasmid of Bacillus
Subtilis, which will secrete it to the culture medium
from where the protein drug will be purified (purification
yet to be tried).
Special Features:
As mentioned above B.subtilis is a
better & safer organism than E.coli. for production
of therapeutic protein. Extracellular secretion by B.subtilis
is an added advantage, as far as purification is concerned.
